Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta.
CD8β chain associates with CD8α to form the CD8αβ heterodimer on thymocytes and cytotoxic T cells.
CD8 receptor binds to MHC class I and is involved in T cell development and activation.
SIDI8BEE detects CD8β in both heterodimer (CD8αβ) and homodimer (CD8ββ) forms.

Applications Reported:

• Flow cytometric analysis.

Applications Tested:

• Pre-diluted and tested by flow cytometry on normal human peripheral blood cells.
• Recommended usage: 5 µL (0.125 µg) per test in 100 µL final volume.
• Suitable cell number: 10⁵–10⁸ cells/test.

Fluorochrome Information:

• PerCP-eFluor 710 emits at 710 nm, excited with blue laser (488 nm).
• Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
• Light-sensitive: protect from light to avoid photo-induced oxidation.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Cat. #00-8222-49)
• 1-step Fix/Lyse Solution (Cat. #00-5333-57)

Target Information

CD8B is a cell surface glycoprotein expressed on most cytotoxic T lymphocytes, mediating cell-cell interactions in the immune system.
Acts as a coreceptor with T-cell receptor recognizing antigens presented by MHC class I molecules.
The functional receptor can be a homodimer (αα) or heterodimer (αβ).
Both chains share homology with immunoglobulin light chains.
The gene encodes CD8β chain isoforms, with multiple alternatively spliced variants.
A pseudogene is also present on chromosome 2.
