Product Specific Information

Description:
The DX2 monoclonal antibody reacts with human CD95 (Fas, Apo-1), a 40–50 kDa member of the TNFR superfamily. CD95 is expressed by a broad range of hematopoietic and non-hematopoietic cells including monocytes, neutrophils, lymphocytes, and fibroblasts. Interaction of CD95 on mature lymphocytes with its ligand (FasL) induces apoptosis and is important in peripheral tolerance. DX2 does not block binding of EOS9.1, another antibody specific for human CD95.

The DX2 monoclonal antibody is also reported to recognize dog/canine CD95.

Applications Reported:
This DX2 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.5 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test).

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm. Ensure your instrument supports detection of this fluorochrome.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FAS (Fas, CD95, APO-1):
A 46 kDa transmembrane glycoprotein functioning as a cell death receptor of the TNFR superfamily. Exists also in a soluble form (~26 kDa). FAS stimulation triggers formation of the death-inducing signaling complex (DISC) and caspase activation, leading to apoptosis. It can also activate NF-κB, MAPK3/ERK1, and MAPK8/JNK pathways, influencing proliferation in fibroblasts and T cells. Multiple splice variants exist, some lacking the transmembrane domain, which may regulate apoptosis negatively. The Fas/FasL system is implicated in diseases such as AIDS, hepatitis, and cancer, and plays a key role in antiviral immune responses.
