Product Specific Information
The W6/32 monoclonal antibody reacts with human MHC class I antigens (HLA-A, B, C). These antigens, associated with beta2-microglobulin, are expressed on all nucleated cells and play a key role in cell-mediated immune response and tumor surveillance.
W6/32 mAb recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity is observed in baboon, rhesus, and cynomolgus monkey.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on human peripheral blood cells at 5 µL (0.6 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample to antibody cocktails prior to combining with cells.
- For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
Technology: NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.
