Product Specific Information

The W6/32 monoclonal antibody reacts with the human major histocompatibility complex (MHC) class I, HLA-A, B, C.
MHC class I antigens associated with beta 2-microglobulin are expressed by all human nucleated cells and play a central role in cell-mediated immune response and tumor surveillance.

W6/32 mAb recognizes a non-polymorphic epitope shared among HLA-A, B, and C molecules and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity is observed in baboon, rhesus, and cynomolgus monkey.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications

• Reported for use in flow cytometric analysis.
• Pre-titrated and tested for flow cytometry of normal human peripheral blood cells.
• Recommended use: 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

HLA-class I major histocompatibility (MHC) antigens are intrinsic membrane glycoproteins expressed on nucleated cells, associated with beta2-microglobulin.
They present foreign peptides for recognition by cytotoxic T cells, playing a key role in antiviral and antitumor defense.
HLA and MHC antibodies are also valuable in Immunopeptidomics, aiding identification of neoantigens via LC-MS/MS.
