Product Specific Information

Description:
The W6/32 monoclonal antibody recognizes human MHC class I antigens (HLA-A, HLA-B, HLA-C) associated with β2-microglobulin. These antigens are expressed by all nucleated human cells and are key components in cell-mediated immune responses and tumor surveillance. The W6/32 clone binds a non-polymorphic epitope common to HLA-A, B, and C loci, and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity has been observed with baboon, rhesus, and cynomolgus monkey.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Tested:

• Pre-titrated and validated for flow cytometry using normal human peripheral blood cells
• Recommended use: 5 µL (0.6 µg) per test (final volume 100 µL)
• Applicable cell number: 10⁵–10⁸ cells/test

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• The product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation and is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample to antibody cocktails before cell staining.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, featuring engineered nucleic acid dye structures that optimize fluorescence signatures and minimize spectral spillover.
