Product Specific Information

The G8.8 monoclonal antibody reacts with the 40 kDa mouse EpCAM (epithelial cellular adhesion molecule), also known as EGP40, 17-1A antigen, TACSTD1, and CD326. Generated using the TE-71 thymic epithelial cell line, CD326 is expressed on most epithelial cells and considered a pan-carcinoma antigen. It mediates calcium-independent, homophilic cell-cell adhesion and may function as a growth factor receptor. CD326 binds LAIR-1 (CD305) and LAIR-2 (CD306), inhibiting activation and inflammation, and plays a key role in tumor biology.

Applications Reported:
This G8.8 antibody is suitable for flow cytometric analysis.

Applications Tested:
Validated in flow cytometric analysis of TE-71 cells. Recommended ≤0.125 µg per test (100 µL final volume). Cell count may vary from 10⁵ to 10⁸ cells/test. Titrate for optimal performance.

Brilliant Violet™ 711 (BV711):
A tandem dye emitting at 713 nm, suitable for cytometers with a 405 nm violet laser.
Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific polymer interactions.
Light-sensitive — protect vial and samples from light.
Not compatible with methanol-based fixation.

Fixation:
Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in dark with minimal brightness loss.

Target Information

Ep-CAM (epithelial adhesion molecule, ESA) is a transmembrane glycoprotein (~40 kDa) expressed in epithelial cells. It functions as a homotypic, Ca²⁺-independent adhesion molecule and influences cell cycle, proliferation, and metabolism via c-myc, cyclin A, and cyclin E induction. Ep-CAM overexpression correlates with enhanced epithelial proliferation and is highly expressed in carcinomas, serving as a marker for epithelial lineage tumors. It is localized on basolateral surfaces of most simple epithelia and carcinoma types, and can distinguish adenocarcinomas from pleural mesotheliomas.
