Product Specific Information

The B2D monoclonal antibody reacts with mouse ROR gamma (t) protein, a member of the retinoic acid-related orphan receptor (ROR) family, which includes ROR alpha and ROR beta. These are ligand-dependent transcription factors involved in multiple physiological processes.

ROR gamma is expressed in tissues such as liver, lung, muscle, heart, and kidney. The isoform ROR gamma t is expressed specifically in lymphoid cells including CD4+CD8+ thymocytes, Th17 cells, and lymphoid tissue inducer (Lti) cells. The B2D antibody recognizes both ROR gamma and ROR gamma t isoforms.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested with mouse splenocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and protocol. Recommended usage ≤ 0.25 µg/test (100 µL final volume).

Fluorophore Information:
PE-eFluor® 610 can be excited with 488–561 nm lasers and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information

ROR gamma (NR1 Thyroid Hormone-Like receptor) influences thymopoiesis, bone metabolism, T-cell apoptosis, and lymphoid organogenesis. It promotes thymocyte survival by activating antiapoptotic protein Bcl-x(L) and is essential for lymph node and Peyer’s patch development.

The thymus-specific isoform ROR gamma(t) inhibits Fas ligand expression and cytokine secretion in immature thymocytes. ROR gamma binds as a monomer to DNA response elements with the GGTCA motif. Expression is documented in mouse thymus, adipose, bone, skeletal muscle, liver, kidney, and human skeletal muscle.
