Product Specific Information

In Western blot, this antibody detects bands at approximately 145 kDa in crude homogenates of rat cerebellum. Human and bovine samples show slower migration at around 160 kDa. A minor band at ~110 kDa may appear, representing an in-vivo proteolytic fragment of NF-M.

Target Information

Neurofilaments (NFs) are intermediate filament proteins expressed almost exclusively in neuronal cells, especially in large axons. They are composed of three major subunits: neurofilament medium (NF-M), high (NF-H), and light (NF-L) proteins. These subunits share sequence and structural similarity in their coiled-coil core domains but differ in their terminal regions, particularly the C-termini of NF-M and NF-H, which form flexible extensions important for filament cross-linking and cytoskeletal architecture.

NF-M plays a key role in maintaining axonal caliber and cytoskeletal structure through its highly phosphorylated tail domain. NF-M phosphorylation and O-GlcNAcylation are reciprocally regulated and influence its translocation, filament formation, and function. Antibodies against neurofilament subunits serve as reliable neuronal markers due to their abundance, neuron-specific expression, and biochemical stability.
