Product Specific Information
For reconstitution, add 100 µL distilled water to obtain a final antibody concentration of about 1 mg/mL.
For conjugation experiments, perform an additional desalting step (Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).
Target Information
Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1.
PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression is upregulated by Th2 cytokine IL-4 and, to a lesser extent, by Th1 cytokines TNF-α and IFN-γ.
PD-L2 has a 2–6 times higher affinity for PD-1 compared to PD-L1, functioning as an inhibitory immune checkpoint molecule that regulates T-cell activation and cytokine secretion.
Recent studies also show PD-L2 binds to a second receptor, RGMb, which plays a role in respiratory tolerance via BMP and neogenin signaling pathways.
The human PD-L2 monomer includes a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
