Product Specific Information

For reconstitution, add 100 µL of distilled water to obtain a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).

Target Information

Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for the programmed death protein 1 (PD-1).
PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Expression is upregulated by cytokines such as IL-4, TNF-α, and IFN-γ.
Although expressed at lower levels than PD-L1, PD-L2 exhibits a 2–6 times higher affinity for PD-1. PD-1 and its ligands function as inhibitory immune checkpoints, providing negative feedback during immune responses.
PD-L2 binding suppresses T-cell activation, proliferation, and cytokine secretion (e.g., IFN-γ, IL-10), but can also stimulate T-cell proliferation through additional receptors such as RGMb.
Interaction between PD-L2 and RGMb regulates respiratory tolerance development in the lung via BMP and/or neogenin signaling pathways.
The human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
