This 4MEJJ monoclonal antibody recognizes mouse aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor mediating responses to environmental contaminants such as PCB and TCDD.
AHR binds to naturally occurring compounds and is localized in the cytoplasm within a complex including HSP90, p23, and XAP2/AIP/ARA9. Upon ligand binding, AHR translocates to the nucleus, associates with ARNT, and binds to DNA sequences regulating genes like CYP1A1.
AHR is highly expressed in liver and plays roles in Treg and Th17 cell differentiation.
The 4MEJJ antibody recognizes b-1, b-2, and d alleles of mouse AHR, but not human AHR.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested in TH17 polarized mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and BestProtocols® Protocol A.

Target Information
AHR (Ah Receptor) is a ligand-activated transcription factor belonging to a family that includes ARNT (HIF-1β) and Drosophila proteins PER and SIM.
It contains an N-terminal PAS domain and regulates genes such as CYPIA1 by binding to enhancer sequences.
AHR controls xenobiotic-metabolizing enzymes like cytochrome P450 and responds to aromatic hydrocarbons including TCDD.
In inactive cells, AHR complexes with HSP90 in the cytoplasm; upon activation, it forms a heterodimer with ARNT for DNA binding.
Diseases associated with AHR include eosinophilic fasciitis and seborrheic dermatitis.
