Description:
This 12.14 monoclonal antibody reacts with mouse Nur77 (also known as NR4A1, TR3, NGFI-B, or NAK1), an inducible orphan nuclear receptor. Nur77 is expressed in thymocytes and T cell lines, promotes apoptosis, and plays a role in thymocyte negative selection. It is also involved in steroid biosynthesis in Leydig cells and dopamine signaling. Additionally, Nur77 interacts with FoxP3 in regulatory T cells, though results with this antibody do not correlate with that observation.
Applications Reported:
This antibody has been reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested:
Tested by intracellular staining and flow cytometry of stimulated mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00).
Recommended use ≤ 1 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test (empirically determined).
Optimize antibody titration for best performance.

Target Information
NR4A1 belongs to the steroid-thyroid hormone-retinoid receptor superfamily. It is expressed in multiple tissues including brain, adrenal, thyroid, liver, testis, ovary, thymus, muscle, lung, and prostate. Expression is induced by stress stimuli and growth factors. The protein functions as a nuclear transcription factor mediating apoptosis and hypothalamic-pituitary signaling. Translocation from nucleus to mitochondria triggers apoptosis. Three transcript variants produce two isoforms, with variant 3 (TRC beta) differing at the C-terminus. RXR is a known partner of NR4A1.
