Target Information

KLRB1 (Killer cell lectin-like receptor subfamily B, member 1, CD161) is a type II membrane protein with an external C terminus. It is expressed by nearly all NK cells and a small subset of CD3+ T cells. KLRB1 forms a homodimeric cell surface protein (40–44 kDa) and plays an inhibitory role on NK cells. Activation of KLRB1 stimulates acid sphingomyelinase/SMPD1 and increases intracellular ceramide, leading to activation of AKT1/PKB and RPS6KA1/RSK1 kinases and T cell proliferation by anti-CD3.

KLRB1 acts as a lectin binding to the Gal-α(1,3)Gal and N-acetyllactosamine epitopes, and also binds CLEC2D/LLT1 as a ligand, inhibiting NK cell-mediated cytotoxicity and interferon-gamma secretion. The protein contains extracellular motifs characteristic of C-type lectins, a transmembrane domain, and a cytoplasmic domain.
