Preparation:
Produced from sera of rabbits immunized with highly pure recombinant murine IL-10. The anti-murine IL-10-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
For detection of mIL-10, use 0.25–1.0 µg/mL of this antibody in conjunction with PeproTech Polyclonal Anti-Murine IL-10 (500-P60) as a capture antibody. Detects at least 0.2–0.4 ng/well of recombinant mIL-10.

Western Blot:
For detection of mIL-10, use 0.1–0.2 µg/mL of this antibody. Detection limit for recombinant mIL-10 is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes, with additional production by lymphocytes. It regulates immune responses and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. IL-10 enhances B cell survival, proliferation, and antibody production, inhibits NF-kappa B activity, and participates in the JAK-STAT signaling pathway. Knockout studies indicate IL-10 is essential for intestinal immune regulation. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis.
