Applications and Tested Dilution

• Flow Cytometry (Flow): 5 µL (0.03 µg)/test
• Tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells.
• A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL.
• Cell number can range from 10⁵ to 10⁸ cells/test.

Product Specific Information

The eBioHP40 antibody reacts with the p40 subunit of human IL-12 p70 and human IL-23.
IL-12 is a 70 kDa heterodimeric cytokine composed of p40 and p35 chains, secreted mainly by activated monocytes, macrophages, and dendritic cells.
IL-23 is a related heterodimeric cytokine composed of disulfide-linked p40 and p19 subunits.

The eBioHP40 antibody shows lower background staining than C8.6 and recognizes a different epitope.

Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for intracellular staining and flow cytometric analysis at 5 µL (0.03 µg) per test.

Fluorochrome Information:
PerCP-eFluor® 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5.
Excitation: 488 nm, Emission: 710 nm, Laser: Blue Laser.
Recommended filter configuration: 685 LP dichroic mirror and 710/40 band pass filter (695/40 also acceptable).

Stability:
Stable when stained samples are exposed to freshly prepared 2% formaldehyde overnight at 4°C.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

IL-12 is produced by dendritic cells, neutrophils, macrophages, and lymphoblastoid cells upon antigenic stimulation.
It is a heterodimeric cytokine encoded by IL-12A (p35) and IL-12B (p40).
IL-23 shares the IL-12B subunit and includes IL-23A (p19).
IL-12 enhances vascular endothelial adhesion molecule-1 expression and facilitates immune cell migration and tumor necrosis.
IL-23 stimulates memory T-cells and is critical for Th17 cell functions, promoting pro-inflammatory cytokine production.
