Product Specific Information

This ebic6 monoclonal antibody reacts with human Epstein-Barr Virus Induced Gene 3 (EBI3), which can be expressed and secreted as monomers or homodimers. EBI3 is a component of two IL-12 family cytokines:

• IL-27 (EBI3 + p28): Secreted by antigen-presenting cells in response to inflammation, with both pro- and anti-inflammatory effects.
• IL-35 (EBI3 + p35): Produced by regulatory T cells, with immunosuppressive functions.

EBI3 is involved in immune regulation, including Th1/Th2/Th17 balance, tolerance during pregnancy, and suppression of effector T-cell proliferation.

• Intracellular staining followed by flow cytometric analysis.

Applications Tested

• Pre-titrated and validated using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824).
• Recommended use: 5 µL (0.03 µg) per test (100 µL reaction volume).
• Suitable for 10⁵–10⁸ cells/test.

• Excitation: 488 nm (Blue laser)

• Stable after fixation with 2% formaldehyde overnight at 4°C.
• 0.2 µm post-manufacturing filtered.

Target Information

EBI3 (Epstein-Barr virus Induced-3) is a secreted glycoprotein of the hematopoietin receptor family.
It regulates Th2-type immune responses and Th2-mediated inflammation, acting through iNKT cell modulation.
EBI3 forms heterodimers:

• IL-27 (EBI3 + p28): Produced by activated antigen-presenting cells; regulates Th17 differentiation.
• IL-35 (EBI3 + p35): Produced by regulatory T cells; suppresses effector T-cell proliferation.

EBI3 expression is controlled by TLR signaling via MyD88 and NF-κB. It may inhibit delayed-type hypersensitivity by suppressing IL-17 and inducing IL-10.
Deficiency in EBI3 is linked to reduced Th2 cytokine production and protection from lung metastasis in mice.
