Description:
The eBioT21/8 monoclonal antibody reacts with the amino terminus of human CCR5 (CD195), an approximately 45 kDa member of the G protein-coupled receptor (GPCR) superfamily that binds chemokines including MIP-1α, MIP-1β, and RANTES. CCR5 is expressed on resting T cells with a memory/effector phenotype, monocytes, macrophages, and immature dendritic cells. It functions as a receptor directing lymphocyte chemotaxis and serves as the primary co-receptor for macrophage-tropic HIV-1, binding through gp120. Binding of this antibody may vary among donors and can be affected by staining conditions.

Applications Tested:
Pre-titrated and validated by flow cytometry on normal human peripheral blood cells. Recommended staining at room temperature or 37°C for 1 hour in Flow Cytometry Staining Buffer (Product # 00-4222-57).

Target Information

HIV and related viruses require co-receptors in addition to CD4 to infect target cells. CCR5, a chemokine receptor, acts as a principal co-receptor for macrophage- and dual-tropic HIV-1 strains, facilitating viral fusion and entry into human white blood cells. CCR5 is essential for infection by HIV-1, HIV-2, and SIV. Its ligands—RANTES, MIP-1α, and MIP-1β—can prevent M-tropic HIV-1 infection. CCR5 mRNA is expressed in lymphoid organs and monocytes, and its amino-terminal domain and extracellular loops serve as HIV binding sites.
