Description:
The NP-6G4 monoclonal antibody reacts with human CD195 (CCR5), an approximately 45 kDa member of the G protein-coupled receptor (GPCR) superfamily that binds chemokines including MIP-1α, MIP-1β, and RANTES. Human CCR5 is expressed on resting T cells with a memory/effector phenotype, monocytes, macrophages, and immature dendritic cells. CCR5 functions as a receptor to direct lymphocyte chemotaxis and acts as a primary co-receptor for macrophage-tropic HIV-1, which binds CCR5 via gp120.

Applications Reported:
This NP-6G4 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
This antibody has been pre-titrated and tested on normal human peripheral blood cells. For optimal performance, staining with NP-6G4 is recommended at 37°C for 1 hour in Flow Cytometry Staining Buffer (Product # 00-4222-57).
Use at 5 µL (0.125 µg) per test. A test is defined as staining a cell sample in 100 µL final volume. Typical cell numbers range from 10⁵ to 10⁸ cells/test.

Target Information

Human immunodeficiency virus (HIV) and related viruses require coreceptors, in addition to CD4, to infect target cells. CCR5, CXCR4, CCR3, CCR2b, and CCR8 are chemokine receptors identified as HIV coreceptors. CCR5 (CC-CKR-5) is the principal coreceptor for macrophage- and dual-tropic HIV-1 strains, facilitating fusion and entry of human white blood cells.
CCR5 is essential for infection by HIV-1, HIV-2, and SIV. Its ligands—RANTES, MIP-1α, and MIP-1β—can prevent M-tropic HIV-1 infection. CCR5 associates with the surface CD4-gp120 complex, enabling membrane fusion and viral entry. The amino-terminal domain and extracellular loops of CCR5 serve as HIV binding sites. CCR5 mRNA is expressed in lymphoid organs and monocytes.
