Antibody detects endogenous levels of total CDC34.

Target Information

CDC34 accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. In vitro, it catalyzes Lys-48–linked polyubiquitination. It cooperates with the E2 UBCH5C and the SCF (FBXW11) E3 ligase complex for the polyubiquitination of NFKBIA, leading to its proteasomal degradation. CDC34 also participates in ubiquitin chain elongation, working with UBE2D3 to build NFKBIA-linked ubiquitin chains. Additionally, it cooperates with the SCF (SKP2) E3 ligase complex to regulate cell proliferation via ubiquitination and degradation of MYBL2 and KIP1. CDC34 is involved in degradation of CREM isoform ICERIIgamma and ATF15, affecting cAMP-induced transcription, and in G2/M phase regulation through WEE1 kinase degradation. It also targets beta-catenin and interacts dynamically with proteasomes under the influence of human herpes virus 1 protein ICP0.
