Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with CD3 gamma and delta, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. SK7 and UCHT1 recognize overlapping epitopes, while OKT3 and SK7 recognize distinct ones. The SK7 antibody also recognizes chimpanzee CD3.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested for flow cytometry using normal human peripheral blood cells. Recommended usage is 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Light Sensitivity: Protect from light; PE-Cyanine5.5 is sensitive to photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information

The CD3 subunit complex is critical for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These subunits associate with intracellular signaling molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. CD3 gene defects are associated with immunodeficiency.
