Product Specific Information
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is required for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Binding of 145-2C11 to TCR induces intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions. Commonly used as a phenotypic marker for mouse T cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.
Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal signal loss.
Excitation/Emission: 488–561 nm / 695 nm
Laser: Blue, Green, or Yellow-Green Laser compatible.
Target Information
The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. CD3 subunits (γ, δ, ε, ζ) are part of the immunoglobulin superfamily encoded on human chromosome 11. These subunits associate with cytoplasmic signaling molecules via a double tyrosine-based motif. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in the CD3 gene are associated with CD3 immunodeficiency.
