Product Specific Information
The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before mixing with cells.
- For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence signatures minimizing spillover and spread.

Target Information
CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 participates in immune responses to bacterial LPS, apoptotic cell binding, and inflammatory processes. Dysregulation is associated with infections (e.g., Mycobacterium chelonae) and respiratory conditions such as croup.
