Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity on monocyte surfaces.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Use 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL of sample (10³–10⁸ cells).

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a receptor for bacterial lipopolysaccharide (LPS) in complex with LBP. Soluble CD14 (sCD14) aids in LPS neutralization and can bind apoptotic cells, influencing immune and inflammatory responses. Dysregulation is associated with infections such as Mycobacterium chelonae and Croup.
