Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest.
Structurally, p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerization domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer agent. It is also linked to aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP).
In normal cells, p53 is expressed at low levels, but in transformed or cancerous cells, it is highly expressed and contributes to malignancy. Mutations in TP53 disrupt DNA binding and abolish tumor suppressor function. Both somatic and germline TP53 mutations are implicated in various cancers, including Li-Fraumeni syndrome.
