Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 is up-regulated under stress conditions and activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 consists of:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer agent. It is also linked to aging, oxidative stress, and cognitive functions such as long-term potentiation (LTP).

In normal cells, p53 levels are low, but in transformed cell lines, expression is high and contributes to malignancy. Mutations in p53 disrupt DNA binding and tumor suppressor activity, and TP53 gene alterations occur in many human cancers and hereditary cancer syndromes such as Li-Fraumeni syndrome.
