Product Specific Information

For reconstitution, add 100 µL of distilled water to obtain a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step (e.g., Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates stress response genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation.
p53 plays a key role in preventing proliferation of damaged cells, acting as an anticancer agent. It is found at low levels in normal cells but elevated in transformed cell lines, contributing to malignancy.
Mutations in TP53 are common in human cancers and can lead to loss of tumor suppressor function. Germline mutations are also linked to cancer-prone syndromes such as Li-Fraumeni syndrome.
Additionally, p53 is associated with aging, oxidative stress, and cognitive function through effects on long-term potentiation (LTP).
