Product Specific Information

The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 plays a key role in peripheral tolerance and autoimmune disease and is expressed mainly on activated T and B lymphocytes. PD-1 ligands include PD-L1 (B7-H1) and PD-L2 (B7-DC), which are expressed on normal and activated antigen-presenting cells.

Costaining experiments indicate that eBioJ105 recognizes a different epitope than MIH4 (Product # 11-9969).

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry on stimulated normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test. One test = staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet 737™ emits at 732 nm and is intended for cytometers equipped with a 355 nm ultraviolet laser. Ensure instrument compatibility.

Staining Buffer Recommendation:
When using polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333). Brilliant Ultra Violet 737™ is compatible with short-term methanol fixation (<1 hour).

Target Information

T-cell activation is regulated by PD-1 (also known as PDCD-1), which binds ligands PD-L1 and PD-L2 to inhibit immune activation in the absence of danger signals. PD-1 deficiency leads to hyperactivated immune responses and autoimmune disease. Despite its predicted molecular weight, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
