Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:

• Flow cytometric analysis.

Applications Tested:

• Pre-titrated and tested on normal human peripheral blood cells.
• Recommended use: 5 µL (0.1 µg) per 100 µL test volume.
• Cell number: 10⁵–10⁸ cells/test (empirically determined).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton technology, based on nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

• Involved in recognition of MHC class II molecules.
• Co-receptor for HIV.
• Expressed in T helper cells, thymocytes, monocytes, macrophages, and dendritic cells.
• Found in thymus, lymph nodes, tonsils, spleen, and some non-lymphoid tissues.
• Functions in T-cell activation via association with the TCR complex and Lck kinase.
• May mediate neuronal damage in infectious and immune-mediated CNS diseases.
• Multiple alternatively spliced transcripts identified (RefSeq, July 2017).
