Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 recognize distinct epitopes.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
This antibody is validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Use at 5 µL (0.1 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures for optimized fluorescence spectra, minimizing spillover and spread.
Excitation: 509 nm; Emission: 665 nm; Laser: 488 nm (Blue).

Target Information

CD4 is a co-receptor for MHC class II molecules and HIV entry. Expressed primarily on T helper cells, monocytes, macrophages, and dendritic cells. Found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 plays a key role in T-cell activation through interaction with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
