Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subpopulation of mature T helper cells, and at low levels on monocytes. CD4 is a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.1 µg)/test. A test is defined as the amount of antibody to stain a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
• For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates are based on Phiton technology utilizing novel nucleic acid dye structures engineered for optimized fluorescence with reduced spillover and spread.

Target Information

CD4 antigen is involved in the recognition of MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed in T helper cells and also found on monocytes, macrophages, and dendritic cells. CD4 participates in T-cell activation via association with the T-cell receptor complex and Lck kinase. It may also mediate neuronal damage in infectious and immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
