Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV; SK3 blocks HIV binding and the mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Use 5 µL (0.1 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample or per 100 µL of cell suspension.
  • Add buffer to antibody cocktails before mixing with cells.

Technology:
NovaFluor conjugates utilize Phiton technology, based on engineered nucleic acid dye structures optimized for minimal spillover and spectral spread.
Excitation: 509 nm; Emission: 614 nm; Laser: 488 nm (Blue).

Target Information

CD4 antigen is involved in MHC class II recognition and functions as a co-receptor for HIV. It is primarily expressed on T helper lymphocytes, as well as monocytes, macrophages, and dendritic cells. CD4 facilitates early T-cell activation via interaction with the T-cell receptor complex and Lck kinase. It may also mediate neuronal damage in infectious and immune-mediated CNS diseases. Expression is observed in thymus, lymph nodes, tonsils, spleen, and specific brain and gut regions. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
