Product Specific Information

Description:
The FNLAP monoclonal antibody reacts with human latency associated peptide (LAP, pro-TGF beta 1, LAP/TGF beta 1). TGF beta is synthesized as a precursor containing LAP at the N-terminus and mature TGF beta at the C-terminus. Cleavage between amino acids 278 and 279 forms LAP and TGF beta dimers, which associate non-covalently to form the small latent TGF beta complex. LAP is secreted into the extracellular matrix and also expressed on platelets and activated regulatory T cells, likely via GARP (LRRC32) binding.

Applications Reported:

  • Flow cytometric analysis.

Applications Tested:

  • Pre-titrated and validated for flow cytometric analysis of stimulated human peripheral blood cells.
  • Recommended use: 5 µL (0.25 µg) per test (100 µL final volume).
  • Cell count range: 10⁵–10⁸ cells/test (empirically determined).

Light Sensitivity:

  • Tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:

  • Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
  • Minimal impact on brightness and FRET efficiency/compensation.
  • Clone-specific performance should be confirmed empirically.

Target Information

TGF beta-1 is a cytokine of the transforming growth factor beta superfamily, found ubiquitously in tissues. It is stored in the extracellular matrix as a latent complex with its prodomain (LAP). Activation requires αV integrin binding to an RGD sequence in the prodomain, enabling release from the extracellular matrix via latent TGF-beta binding proteins (LTBP). The latent complex includes TGF-beta, LAP, and LTBP.
