Product Specific Information

The A3KCAT antibody recognizes human MerTK (also known as MER or c-mer), a 170–210 kDa member of the TAM family of tyrosine kinase receptors, which includes Axl and Tyro3. MerTK is expressed on a subset of anti-inflammatory macrophages and plays a key role in the removal of apoptotic cells through binding with Protein S and Gas6 ligands. Upon ligand binding, MerTK undergoes autophosphorylation, activating the PI3K and Akt pathways, leading to phagocytosis and suppression of pro-inflammatory cytokine production.

MerTK deficiency can result in autoimmunity and inflammation, and its expression is often observed in malignant cells, suggesting a potential role in immune evasion. This antibody has been validated in IHC of formalin-fixed paraffin-embedded tissue using both low and high pH antigen retrieval. Recommended working concentration is ≤0.5 µg/mL, with titration advised for optimal assay performance.

Target Information

MERTK (cMER) is a tyrosine kinase proto-oncogene involved in retinal pigment epithelium (RPE) phagocytosis and other physiological processes including cell survival, migration, differentiation, and efferocytosis. Ligand binding induces autophosphorylation and downstream signaling via GRB2 or PLCG2, leading to phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1. MERTK signaling contributes to macrophage clearance of apoptotic cells, platelet aggregation, cytoskeletal reorganization, and inhibition of TLR-mediated innate immune responses through STAT1 activation and induction of SOCS1/SOCS3.
