Product Specific Information

Monoclonal antibody J5HTR3 reacts with human CD204, also known as Macrophage Scavenger Receptor 1 (MSR1), Scavenger Receptor Type A (SR-A), or SCARA1.
CD204 is expressed by multiple macrophage and dendritic cell populations and belongs to the class A family of scavenger receptors, which includes MARCO (SCARA2), CSR (SCARA3), SRCL (SCARA4), and SCARA5.

CD204 binds and internalizes oxidized LDL, contributing to macrophage differentiation into foam cells, secretion of inflammatory cytokines, and development of atherosclerotic lesions.
It also uptakes various macromolecules such as bacterial cell wall components, viral DNA, calciprotein complexes, apoptotic cells, and beta-amyloid fibrils.
These interactions link CD204 to processes including host defense, inflammation, bone remodeling, atherosclerosis, sepsis, and Alzheimer’s disease.

Isoforms

Human CD204 exists as three splice variants:

  1. Type I (SR-AI): Full-length, ubiquitous isoform
  2. Type II (SR-AII): Lacks C-terminal cysteine-rich domain
  3. Type III (SR-AIII): Shortest, not expressed on cell surface

J5HTR3 binds both Type I and Type II isoforms (Type III not tested).

Tested by immunohistochemical staining of formalin-fixed paraffin-embedded human tissue using high pH antigen retrieval.
Recommended concentration ≤ 0.5 µg/mL; titrate for optimal performance.


Target Information

The CD204 (MSR1) gene encodes class A macrophage scavenger receptors, including three isoforms (1, 2, 3) generated by alternative splicing.
These trimeric integral membrane glycoproteins are macrophage-specific and implicated in physiological and pathological processes such as atherosclerosis, Alzheimer’s disease, and host defense.

  • Isoforms 1 and 2: Functional receptors mediating endocytosis of modified LDLs
  • Isoform 3: Retained in endoplasmic reticulum, non-functional
  • Isoform 3 may inhibit isoforms 1 and 2 when co-expressed (dominant negative effect)

Diseases associated with MSR1 include Barrett Esophagus and Prostate Cancer.
