Product Specific Information

This 10H11.E12 monoclonal antibody recognizes human, mouse, and rat ataxia-telangiectasia mutated (ATM) protein phosphorylated at serine 1981. ATM is a PI3 kinase family member that coordinates cell cycle arrest and DNA repair initiation through phosphorylation of multiple substrates in response to DNA damage and oxidative stress.

ATM exists as inactive dimers or multimers but becomes activated by monomer formation and autophosphorylation upon DNA double-strand breaks. Activated ATM halts the cell cycle and initiates DNA repair. Loss of ATM function leads to ataxia-telangiectasia (AT), characterized by neurodegeneration, immunodeficiency, radiosensitivity, and cancer predisposition (e.g., lymphoma, leukemia).

Applications Reported:
Suitable for intracellular staining followed by flow cytometry, immunoprecipitation, western blotting, immunocytochemistry, and immunohistochemistry of formalin-fixed paraffin-embedded tissues. Fluorochrome-conjugated 10H11.E12 is recommended for intracellular flow cytometry.

Applications Tested:

  • IHC (paraffin): ≤10 µg/mL (high or low pH antigen retrieval)
  • ICC: ≤10 µg/mL (methanol or formaldehyde-fixed cells)
    Careful titration is recommended for optimal assay performance.

Target Information

Ataxia-telangiectasia Mutated (ATM) is a PI3/PI4 kinase family protein. The ATM gene encodes a kinase that phosphorylates numerous downstream targets, including P53, Mdm2, BRCA1, and SMC1, regulating cell cycle and DNA repair. ATM is activated by autophosphorylation following DNA damage such as ionizing radiation. Deficiency in ATM results in hypersensitivity to radiation, impaired DNA synthesis inhibition, delayed p53 induction, and dysregulated apoptosis via the E2F1 pathway involving Nbs1 and Chk2.
