Product Specific Information

The 2F10 monoclonal antibody recognizes human osteopontin (OPN), a secreted glycophosphoprotein expressed by various cell types such as leukocytes, fibroblasts, dendritic cells, macrophages, myoblasts, bone-forming cells, and epithelial cells. OPN functions as an extracellular matrix protein involved in bone resorption and calcification, immune regulation, wound healing, and tumorigenesis. Thrombin cleavage of OPN enables integrin receptor binding, influencing cell adhesion, migration, and survival. Expression of OPN is found in bones, teeth, placenta, blood vessels, kidney, and numerous tumors. It is considered a potential therapeutic target for autoimmune diseases, cancer metastasis, bone mineralization disorders, and osteoporosis.

Applications Reported:
Validated for Western blotting, immunohistochemistry of formalin-fixed paraffin-embedded tissue, and immunocytochemistry.

Applications Tested:

• IHC (P): ≤5 µg/mL (using low or high pH antigen retrieval)
• ICC/IF: ≤10 µg/mL (methanol-fixed or formaldehyde-fixed and permeabilized cells)
  Antibody should be titrated for optimal performance.

Target Information

Osteopontin is an RGD-containing glycoprotein that interacts with integrins and CD44 as major receptors. It plays a role in bone mineralization, cell adhesion, migration, chronic inflammation, and cellular transformation. Proteolytic cleavage by thrombin and matrix metalloproteinases near the integrin-binding site modulates its function and binding properties. Thrombin-cleaved fragments are overexpressed in malignant glial tumors, providing survival advantages and serving as substrates for plasmin and cathepsin D.
