Description:
The 25-D1.16 monoclonal antibody recognizes the ovalbumin-derived peptide SIINFEKL bound to H-2Kb of MHC class I, but not unbound H-2Kb or irrelevant peptide complexes. It is useful for tracking the quantity and localization of specific antigen-presenting cells (APC) in vivo.

Applications Reported:
This eBio25-D1.16 (25-D1.16) antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of SIINFEKL-pulsed C57Bl/6 splenocytes. Recommended ≤0.125 µg per test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.
