Product Specific Information
- For Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
Neuron Specific Enolase (NSE, ENO1, ENO2, ENO3) is an enzyme that catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate in glycolysis and the reverse reaction in gluconeogenesis. NSE is stable in biological fluids and can diffuse into extracellular media and cerebrospinal fluid when neuronal membranes are injured.
NSE exists as three mammalian isoforms (α, β, γ).
- α-subunit: expressed in most tissues
- β-subunit: expressed in muscle
- γ-subunit: expressed primarily in neurons and neuroendocrine cells
Co-expression of NSE and chromogranin A is common in neuroendocrine neoplasms. Antibodies to NSE are useful for identifying neuronal cell bodies, developing neuronal lineage, and neuroendocrine cells. Elevated NSE levels are biomarkers of neuronal injury and are clinically used to assess damage in conditions such as stroke, hypoxic brain injury, status epilepticus, Creutzfeldt-Jakob disease, and herpetic encephalitis. NSE is also elevated in certain neoplasms including pediatric neuroblastoma and small cell lung cancer.
