Target Information
Protein p21 is a tumor suppressor protein that inhibits cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and regulates cell cycle progression by blocking transitions from G1 to S phase or G2 to mitosis after DNA damage.
In the nucleus, p21 binds and inhibits Cdk1 and Cdk2, facilitating DNA repair. In the cytoplasm, p21 exhibits anti-apoptotic effects by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK.
P21 has dual roles in carcinogenesis, acting both as a tumor suppressor and as an oncogene depending on cellular context.
