Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα).

• Expression: Thymocytes, mature αβ T cells, γδ T cells, IELs, dendritic cells
• Function: CD8 binds to MHC class I and associates with p56lck kinase, contributing to T cell development and activation.
• Applications Reported: Flow cytometric analysis of mouse splenocytes
• Recommended Use: ≤ 0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

Each NovaFluor conjugate or kit ships with CellBlox Blocking Buffer.

• Add 5 µL per sample when staining with NovaFluor conjugates.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein functioning as a co-receptor with TCR for MHC class I molecules.

• Expression: T cells, NK cells, monocytes, dendritic cells
• Structure: Disulfide-linked heterodimer (CD8αβ) or homodimer (CD8αα)
• Function: Mediates signal transduction, T cell differentiation, and activation.
• Activation Pathway: MHC-I/peptide binding triggers Lck recruitment, leading to cytokine production and cytotoxic T lymphocyte activation.
• Regulation: Enhanced by IL-2, IL-12, and TGF-β1.
