Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. CD8 is expressed on most thymocytes and subsets of mature T cells, IELs, and dendritic cells, playing a key role in T cell development and activation through MHC class I binding and p56lck association.

Applications

• Reported for flow cytometric analysis of mouse splenocytes
• Recommended ≤ 0.5 µg/test in 100 µL final volume (10⁵–10⁸ cells/test)
• Optimize titration for best performance

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Always include CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) in antibody cocktails for optimal staining.
  • Add 5 µL per sample (regardless of number of NovaFluors).
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein in the immunoglobulin receptor family. It functions as a co-receptor with the TCR for MHC class I molecules, facilitating T cell activation and differentiation. CD8 is mainly expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells. Engagement with MHC-I/peptide complexes recruits Lck kinase, promoting cytokine production and cytotoxic T lymphocyte activation. CD8+ T cell differentiation into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
