Product Specific Information

The eBioH4A3 monoclonal antibody reacts with human CD107a (LAMP-1), a highly glycosylated protein (~110 kDa) primarily located in lysosomal/endosomal membranes. It transiently appears on the surface of degranulating cytolytic T cells and is upregulated on activated platelets and some cancer cells.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of Jurkat cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).

  • Recommended use: 5 µL (0.25 µg) per test in 100 µL total volume.
  • Suitable for 10⁵–10⁸ cells/test (optimize empirically).
  • Excitation: 488–561 nm; Emission: 607 nm.
  • Recommended filter: 610/20 band pass (equivalent to PE-Texas Red®).
  • Light sensitive: protect from light.
  • Fixation: Samples can be stored in IC Fixation Buffer (#00-8222) or 1-step Fix/Lyse Solution (#00-5333) for up to 3 days at 4°C in the dark.
  • Filtered through 0.2 µm post-manufacturing.

Target Information

LAMP1 (CD107a) is a major lysosomal membrane protein, together with LAMP2, comprising a significant portion of the lysosomal membrane. It plays a key role in lysosome biogenesis and fusion with phagosomes. LAMP1 is a type I integral membrane protein with 18 N-linked glycosylation sites and a short cytosolic tail.

  • Involved in lysosome–endosome transport and plasma membrane translocation upon activation.
  • Surface LAMP1 promotes PBMC adhesion to vascular endothelium, suggesting a role in inflammation.
  • Increased expression observed in neurons and glial cells near senile plaques in Alzheimer’s disease.
  • Marker of mast cell activation.
