MN1060 recognized PHF-Tau and shows no cross-reactivity with normal or fetal Tau. This indicates detection of an abnormal phosphorylation site unique to PHF-Tau.
The epitope contains phosphorylated Ser214 and Thr212 residues (numbering according to human Tau40).
MN1060 detects PHF-Tau (Thr212/Ser214) with a predicted molecular weight of approximately 79 kDa.
This product is a Low Endotoxin formulation.
Purity >95% as determined by SDS-PAGE.

Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules.
The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, functioning as a linker protein.
Axonal polarity is determined by TAU/MAPT localization.
Short isoforms contribute to cytoskeletal plasticity; longer isoforms aid stabilization.
In its hyper-phosphorylated form, Tau is a major component of paired helical filaments (PHF), the building block of neurofibrillary lesions in Alzheimer’s disease (AD).
Hyper-phosphorylation impairs microtubule binding, destabilizing microtubules and leading to neuronal degeneration.
Tau is phosphorylated by multiple kinases including GSK-3β, PKA, cdk5, and casein kinase II.
Hyper-phosphorylated Tau is also found in other CNS disorders such as Pick’s disease, frontotemporal dementia, cortico-basal degeneration, and progressive supranuclear palsy.
