Product Specific Information
MN1050 recognizes PHF-Tau, tangles, and neurofilaments. It cross-reacts weakly with normal Tau and does not cross-react with recombinant unphosphorylated Tau. The epitope corresponds to the phosphorylated Thr181 residue (numbering according to human Tau40).
MN1050 detects PHF-Tau (Thr181), with a predicted molecular weight of approximately 79 kDa.
Purity is >95% as determined by SDS-PAGE.
Target Information
Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus binds neural plasma membrane components, functioning as a linker between both structures.
Axonal polarity is determined by TAU/MAPT localization within the neuronal cell body domain defined by the centrosome. Short isoforms contribute to cytoskeletal plasticity, whereas longer isoforms stabilize the structure.
In its hyper-phosphorylated form, Tau forms paired helical filaments (PHF), the primary component of neurofibrillary tangles in Alzheimer’s disease (AD) brains. Hyper-phosphorylation disrupts Tau’s microtubule-binding ability, leading to microtubule destabilization and neuronal degeneration in AD.
Tau phosphorylation is mediated by several kinases, including GSK-3β, PKA, cdk5, and casein kinase II. Hyper-phosphorylated Tau is also found in neurofibrillary lesions of other CNS disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
