Target Information
Estrogen Receptor beta (ER beta, NR3A2) is a nuclear receptor protein (~55 kDa) with multiple isoforms (36–59 kDa). Initially characterized from human testis, ER beta is part of the steroid/thyroid hormone receptor superfamily. Upon estrogen binding, ER beta undergoes conformational change, dimerizes, and binds to estrogen response elements (EREs) in DNA, regulating transcription through coactivator or corepressor recruitment.
Two forms of estrogen receptors exist—alpha (ESR1) and beta (ESR2). ER beta binds estrogen with similar affinity to ER alpha and may inhibit alpha receptor activity in reproductive tissues. ER beta is widely expressed and acts as a tumor suppressor, implicated in diseases such as prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain Disease, and cardiovascular disease risk.
