Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature. Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type 1 glycoprotein with a molecular weight of 40–43 kDa. It is expressed in many tumors and normal cells, particularly lymphatic epithelial cells and follicular dendritic cells. Podoplanin localizes in stromal cells of peripheral lymphoid tissue and thymic epithelial cells.

As a regulator of lymphatic endothelium, podoplanin likely contributes to maintaining the unique shape of podocytes. It serves as a ligand for CLEC-2, and its expression correlates with tumor invasiveness and metastatic potential. Podoplanin is involved in cell migration, metastasis formation, and tumor invasion.

Podoplanin expression can be induced via Src activation of Cas, and recent studies indicate its presence on Th17 cells. Initial research has shown podoplanin on a distinct population of CD4+ cells from human PBMC. Dysfunction of this protein is linked to diseases such as lymphangioma and subglottic neoplasm. It has also been proposed as a marker for lung injury due to its physiological role.
