Product Specific Information

  • Antibody is tested in direct ELISA.
  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein that acts as a cell death receptor of the TNFR (tumor necrosis factor receptor) superfamily. It also exists in a soluble form (~26 kDa).
Upon stimulation, FAS aggregates its intracellular FADD domains, forming the death-inducing signaling complex (DISC) and activating caspases. This cascade leads to apoptosis.
FAS also activates NF-kappaB, MAPK3/ERK1, and MAPK8/JNK, contributing to proliferative signaling in fibroblasts and T cells.
Multiple alternatively spliced isoforms exist, with some lacking the transmembrane domain, which may negatively regulate apoptosis.
The Fas/Fas ligand system plays roles in diseases such as AIDS, hepatitis, and cancer, and is important in antiviral immune responses.
