Product Specific Information

  • Antibody is tested in direct ELISA.
  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

FAS (Fas, CD95, APO-1) is a 46 kDa transmembrane glycoprotein acting as a cell death receptor of the TNFR (tumor necrosis factor receptor) superfamily. It also exists in a soluble form (~26 kDa).
Stimulation of FAS leads to aggregation of FAS-associated death domains (FADD), formation of the death-inducing signaling complex (DISC), and activation of caspases.
Caspase activation triggers apoptosis via downstream signaling cascades.
FAS can also activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK, and is involved in proliferative signaling in fibroblasts and T cells.
Multiple alternatively spliced variants exist, with some lacking the transmembrane domain, potentially regulating apoptosis negatively.
The Fas/Fas ligand system is implicated in diseases such as AIDS, hepatitis, and cancer, and is key in antiviral immune responses.
