Product Specific Information

Alexa Fluor™ Plus recombinant antibodies utilize proprietary dye chemistry for enhanced brightness and improved signal-to-noise ratio compared to conventional Alexa Fluor conjugates. These recombinant antibodies offer superior specificity and lot-to-lot consistency by expressing cloned antibody genes in vitro.

Usage Note:
Before use, centrifuge the conjugate solution briefly and use only the supernatant to minimize nonspecific background staining caused by aggregates.

Tested Applications:
This OC-3F10 antibody has been validated for immunocytochemistry and flow cytometry using Caco-2 cells.

• ICC: 1 µg/mL
• Flow Cytometry: ≤ 0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
  Optimal dilution should be determined experimentally.

Target Information

Occludin is a 65 kDa protein that can appear in multiple phosphorylated forms up to ~82 kDa. It plays a key role in tight junction integrity and permeability regulation. Polyunsaturated fatty acids up-regulate occludin expression, enhancing cell resistance and decreasing permeability. Expression levels vary by tissue, with high and continuous expression in brain endothelial cells. Reduced occludin expression is associated with increased tumor endothelial permeability and brain tumor edema.
