Product Specific Information

The monoclonal antibody B34D1.3 recognizes human podocalyxin, a protein expressed on undifferentiated human embryonic stem cells (ES), embryonic carcinoma cells (EC), and embryonic germ cells (EG). Podocalyxin is a heavily glycosylated cell adhesion protein with homology to CD34 and endoglycan. It was first identified on kidney podocytes and later found to be upregulated in various human cancers such as breast and prostate cancer. The epitope recognized by B34D1.3 is lost upon cell differentiation.

B34D1.3 recognizes the ectodomain of podocalyxin, distinct from the epitopes recognized by Tra-1-60 and Tra-1-81 antibodies. Tra-1-60 reacts with a sialidase-sensitive epitope, while the Tra-1-81 epitope remains undefined.

Target Information

This gene encodes a member of the sialomucin protein family, originally identified as an important component of glomerular podocytes. Podocytes are highly differentiated epithelial cells with interdigitating foot processes covering the outer aspect of the glomerular basement membrane.

Additional biological activities include:

  • Binding in a membrane protein complex with Na⁺/H⁺ exchanger regulatory factor to cytoskeletal elements
  • Playing a role in hematopoietic cell differentiation
  • Expression in vascular endothelium and binding to L-selectin
